QJO1MA01 - Ofloxacin. Ofloxacin is an antibiotic from the fluoroquinolone group that is active against Gram-negative microorganisms, namely: Esherichia coli, Salmonella spp, Proteus spp, Shigella spp, Klebsiella spp, Enterobacter spp, Camphylobacter spp, Yersinia spp, Pseudomonas aeruginosa, Vibrio spp, Aeromonas spp, Haemophilus spp, Serratia spp, Chlamydia spp. Active against some Gram-positive microorganisms, in particular Staphylococcus spp. (including penicillinase-producing and methicillin-resistant strains), Streptococcus spp. Moderately sensitive to ofloxacin are Enterococcus faecalis, Pseudomonas spp. Also sensitive to the drug are: Brucella spp. The mechanism of action of ofloxacin is to inhibit bacterial DNA gyrase (topoisomerase II), which stops the process of spiralization and DNA synthesis. A significant advantage of the drug compared to other topical antibiotics is its high permeability to the cornea and anterior chamber of the eye. Among all fluoroquinolones, ofloxacin has the best ability to penetrate the cornea and anterior chamber of the eye, its effective concentration in the tear film remains even 4 hours (240 minutes) after administration, and its MIC90 is above 2 μg/ml. Systemic reabsorption after topical administration into the conjunctival sac is insignificant and has no clinical significance. Instillation of ofloxacin into the ear with an intact eardrum is accompanied by minimal absorption in the middle ear. However, the absorption of ofloxacin increases in cases of perforation of the tympanic membrane. After instillation of ofloxacin in adult animals with tympanic membrane perforation, the maximum serum concentration is 10 ng/ml. After topical administration, therapeutic concentrations of ofloxacin are achieved in the eye tissues within 30-60 minutes. After 10 days of administration of the drug in therapeutic doses, the concentration of ofloxacin in blood serum was 1000 times lower than the standard concentration after oral administration. 🟡STORAGE: Store in a dry, dark place out of reach of children and animals, away from food and feed, at a temperature of 5 to 25°C. 🟡DURABILITY: 3 years. After first withdrawal from the vial, the medicine should be used within 30 days. For use in veterinary medicine!
